Proper reconstitution is essential for maintaining peptide integrity and ensuring reliable results. This guide walks you through the standard process for reconstituting lyophilized (freeze-dried) peptides with bacteriostatic water or other appropriate solvents.

What You’ll Need

Before beginning the reconstitution process, gather the following supplies:

  • Lyophilized peptide vial — your NPepSpacetides product in powder form
  • Bacteriostatic water (BAC water) — sterile water containing 0.9% benzyl alcohol as a preservative
  • Insulin syringes — for precise volume measurement (1mL / 100 unit syringes recommended)
  • Alcohol swabs — for sterilizing vial stoppers before each use
  • Clean workspace — a sanitized, dust-free surface for preparation
  • Gloves — nitrile or latex gloves to maintain sterility

Step-by-Step Reconstitution Process

Step 1: Prepare Your Workspace

Clean your work surface thoroughly with isopropyl alcohol or a suitable disinfectant. Wash your hands and put on clean gloves. Lay out all supplies within easy reach. A clean environment minimizes the risk of contamination.

Step 2: Allow the Peptide to Reach Room Temperature

Remove the peptide vial from cold storage and allow it to reach room temperature naturally. This typically takes 15–20 minutes. Do not heat the vial or attempt to speed up this process — rapid temperature changes can damage the peptide. Allowing the vial to warm prevents condensation from forming inside the vial when it’s opened.

Step 3: Sterilize the Vial Stoppers

Using an alcohol swab, thoroughly wipe the rubber stopper on top of the peptide vial. Do the same for the bacteriostatic water vial. Allow the alcohol to air dry completely before proceeding — this takes about 30 seconds. This step prevents microbial contamination.

Step 4: Draw the Bacteriostatic Water

Using a clean insulin syringe, draw the desired amount of bacteriostatic water from the BAC water vial. The volume you use depends on the peptide amount and your desired concentration. A common starting point is 1 mL of BAC water per 5 mg of peptide, but adjust based on your protocol requirements.

Volume Tip: Adding more solvent creates a lower concentration (easier to measure small amounts), while less solvent creates a higher concentration (fewer injections for larger amounts). Calculate your desired concentration before adding solvent, as the process is not easily reversible.

Step 5: Add Water to the Peptide Vial

Insert the syringe needle through the rubber stopper of the peptide vial. Do not inject the water directly onto the powder. Instead, angle the needle so the water runs down the inside wall of the vial. Add the water slowly and gently — rushing this step can damage the peptide structure.

Key points:

  • Let the water slide down the glass wall, not blast onto the powder
  • Add the water slowly over 30–60 seconds
  • Do not shake, vortex, or agitate the vial vigorously

Step 6: Allow the Peptide to Dissolve

After adding the water, gently swirl the vial in a slow circular motion to encourage dissolution. Never shake the vial. Most peptides will dissolve completely within 5–10 minutes. If the powder hasn’t fully dissolved, let it sit at room temperature and check again in a few minutes. The solution should be clear and free of visible particles when fully reconstituted.


Storage After Reconstitution

Proper storage of reconstituted peptides is critical for maintaining potency and stability:

  • Refrigerate immediately: Store reconstituted peptides at 2–8°C (standard refrigerator temperature)
  • Use within the recommended timeframe: Most reconstituted peptides remain stable for 3–4 weeks when refrigerated with bacteriostatic water. Check your product’s specific documentation for exact timeframes
  • Avoid freezing reconstituted solutions: Freezing and thawing can cause peptide degradation and loss of potency
  • Keep away from light: Store in the original vial and keep away from direct light exposure
  • Always sterilize the stopper: Use a fresh alcohol swab before each withdrawal from the vial

Why Bacteriostatic Water? Bacteriostatic water contains 0.9% benzyl alcohol, which inhibits bacterial growth and extends the usable life of your reconstituted peptide. If you use plain sterile water instead, the solution must be used within 48–72 hours due to the lack of preservative.

Safety Guidelines

  • Always wear gloves and work in a clean environment
  • Never share syringes or needles between vials
  • Dispose of used needles and syringes in an appropriate sharps container
  • If the reconstituted solution appears cloudy, discolored, or contains particles, do not use it — contact our support team
  • Keep all peptides and supplies out of reach of children and unauthorized personnel
  • Follow your institution’s biosafety and waste disposal protocols

Common Mistakes to Avoid

Injecting Water Directly onto the Powder

Blasting water directly onto the lyophilized cake can damage the peptide’s molecular structure. Always aim the needle at the vial wall and let the water trickle down gently.

Shaking the Vial

Vigorous shaking creates foam and can denature the peptide through excessive agitation. Use slow, gentle swirling motions instead.

Using Too Little or Too Much Solvent

Adding too little water makes measurement difficult and increases the risk of dosing errors. Adding too much dilutes the solution excessively. Calculate your desired concentration before you begin.

Skipping the Warm-Up Step

Reconstituting a cold vial can cause condensation and temperature shock. Always let the vial reach room temperature before adding solvent.

Not Sterilizing Vial Stoppers

Skipping the alcohol swab step introduces contamination risk. Clean the stopper every single time, even if you just cleaned it minutes ago.

Storing Reconstituted Peptides at Room Temperature

Reconstituted peptides degrade rapidly at room temperature. Refrigerate immediately after reconstitution and between uses.
